One-Pot Bi-Enzymatic Cascade Synthesis of Novel Ganoderma Triterpenoid Saponins

Ganoderma lucidum is a medicinal fungus whose numerous triterpenoids are its main bioactive constituents. Although hundreds of have been identified, triterpenoid glycosides, also named saponins, rarely found. Ganoderic acid A (GAA), major triterpenoid, was synthetically cascaded to form GAA-15-O-?-glucopyranoside (GAA-15-G) by glycosyltransferase (BtGT_16345) from Bacillus thuringiensis GA A07 and subsequently biotransformed into series GAA glucosides cyclodextrin glucanotransferase (Toruzyme® 3.0 L) Thermoanaerobacter sp. The optimal reaction conditions for the second-step biotransformation GAA-15-G were found be 20% maltose; pH 5; 60 °C. (GAA-G2, GAA-G3, GAA-G4) could purified with preparative high-performance liquid chromatography (HPLC) identified mass nucleic magnetic resonance (NMR) spectral analysis. product, GAA-15-O-[?-glucopyranosyl-(1?4)-?-glucopyranoside] (GAA-G2), showed over 4554-fold higher aqueous solubility than GAA. present study demonstrated that multiple saponins produced sequential actions BtGT_16345 Toruzyme®, synthetic strategy we proposed might applied many other produce novel in future.
